10 Smallpox Remains a Significant Threat Today Current geopolitical environment heightens the need for vigilance The Growing Threat 30% Historic mortality rate More than 300M Estimated deaths in the 20th century alone from smallpox Smallpox continues to pose a serious global threat. Smallpox can spread globally before symptoms appear, leading to catastrophic impact. The ongoing geopolitical issues and advancements in synthetic biology are increasing the likelihood of potential threats. Vaccines alone may not be enough Comprehensive Containment Measures are Critical Global travel increases the potential exposure facilitating the rapid spread across borders Most of today’s population is not vaccinated against smallpox, leaving millions vulnerable to its spread. CDC Model of a 33% transmission reduction Quarantines and vaccines alone may not be enough to stop the spread of smallpox within one year. START DAY 45 START DAY 30 START DAY 25 DAYS POST RELEASE TOTAL CASES (Thousands) 1 50 99 148 197 246 295 344 Result: No Control Vaccines and antivirals are both needed in a comprehensive biodefense preparedness plan, safeguarding vulnerable populations against smallpox threats.

12 As the first FDA-approved antiviral treatment for smallpox, TPOXX was among the first therapies delivered to the U.S. SNS under Project BioShield. TPOXX’s was developed in partnership with, and with funding from, BARDA. Prevents the variola virus, which causes smallpox, from leaving infected cells This slows the spread of the infection allowing the immune system to clear the virus Has a strong safety profile and is well-tolerated in patients TPOXX: The Essentials Strong Preclinical and Clinical Data Preclinical Data - Significantly reduced both mortality and viral load in 4 pivotal studies in non-human primates and 2 pivotal studies in rabbits. Clinical Data - Safety demonstrated in 17 clinical trials involving administration in 1,300+ healthy human volunteers - Studies provided quantifiable pharmacokinetic (PK) data within efficacious dose ranges and resulted in no serious adverse events. 2018 U.S. FDA 2021 Health Canada 2022 EMA and UK MHRA 2024 Japan PMDA How it works Approvals A highly targeted antiviral treatment against orthopoxviruses, including smallpox

Health Security Gap 15 TPOXX as a PEP Aims to Expand Health Benefits When Exposed to Smallpox Totality of data suggests administration is optimal as early as possible or as post-exposure prophylaxis FeverIncubation (Asymptomatic Stage) Antiviral Post-Exposure Prophylaxis Antiviral Treatment for Active Infections Scabs and Scarring Day 1 Day 3 Day 12 Day 14 Day 30 Rash Infectious Stage Exposure Vaccines Window Optimally within 3 days but no later than 7 days Note: TPOXX is not approved as a post-exposure prophylaxis in any jurisdiction. The current TPOXX indication leaves a health security gap between the time when vaccines cease to be effective and the onset of symptoms, triggering therapeutic treatment An expanded label to include PEP would address this gap Nonclinical studies have shown that early administration of TPOXX after exposure leads to improved outcomes
